Depemokimab is associated with early and sustained improvements in patient-reported outcomes in CRSwNP: Data from the ANCHOR-1/2 trials

Background: Depemokimab is the first ultra-long-acting biologic with enhanced IL-5 binding affinity, high potency and an extended half-life, enabling sustained suppression of type 2 inflammation and twice-yearly dosing. Aims: Investigate the earliest impact to patient-reported outcomes (PROs) with depemokimab in the integrated population of the Phase III ANCHOR-1/2 trials. Methods: Adults with inadequately controlled CRSwNP were randomised (1:1) to depemokimab (100 mg subcutaneous; n=272) or placebo (n=256), plus standard of care, once every 26 weeks for 52 weeks. Symptom-specific verbal response scales (VRS) for nasal obstruction and loss of smell (scale 0–3; higher scores indicate worse outcomes) were self-assessed daily and a mean weekly score was calculated per symptom. Results: Improvements in change from baseline in mean nasal obstruction VRS were observed by Week 3 with depemokimab (difference vs placebo [95% CI]: –0.15 [–0.29,–0.02], p=0.028) and by Week 4 for loss of smell VRS (–0.15 [–0.26,–0.03], p=0.012; both nominally significant). Improvements remained nominally significant at each timepoint until the end of the study (Week 52; Figure). Conclusions: Twice-yearly depemokimab provided early and sustained improvements in nasal obstruction and loss of smell, two cardinal and burdensome symptoms of severe CRSwNP.
