\nCacar monyet merupakan penyakit zoonis yang disebabkan oleh virus Monkeypox. Kasus infeksi virus Monkeypox pada manusia sejak tanggal 6 Mei 2022 telah dilaporkan teridentifikasi di Britania Raya, kemudian diikuti oleh beberapa negara seperti Spanyol, Portugal, Jerman, Perancis, Canada, Belanda, hingga Indonesia. World Healt Organization (WHO) telah menetapkan kasus infeksi virus Monkeypox sebagai darurat kesehatan global. Virus Monkeypox menginfeksi manusia dalam bentuk struktur Intracellular Mature Virion (IMV) melalui protein membran yakni protein E8L. Protein E8L IMV memiliki daerah yang dapat dikenali dan berikatan dengan antibodi, disebut sebagai epitop sel B. Epitop sel B telah banyak dikaji untuk menemukan susunan peptida sebagai bahan vaksin berbasis peptida. Tujuan dari penelitian ini adalah untuk menemukan sekuen epitop sel B potensial untuk kandidat vaksin berbasis peptida. Penelitian ini dilakukan secara in silico melalui pemetaan epitop sel B, kemudian beberapa kandidat epitop diuji berdasarkan beberapa parameter seperti uji antigenitas, uji alerginisitas, uji hidrofobisatas, simulasi docking, dan analisis interaksi molekular. Hasil penelitian ini adalah epitop sel B potensial untuk kandidat vaksin peptida berdasarkan uji antigenitas (0.4274), alergenitas, hidrofobisitas (-2.418), dan energi ikatan hasil interkasi docking (-274.0) adalah epitop dengan sekuen KKKYSSYYEEAKK. Interaksi molekular antara epitop potensial dengan reseptor sel B (ID. 5IFH) adalah melalui asam amino lysine 1, lysine 2, dan tyrosin 4.

\nMonkeypox is a zoonotic disease caused by the Monkeypox virus. Viral infection case Monkeypox in humans since May 6 202has been reported identified in the United Kingdom, followed by several other countries such as Spain, Portugal, Germany, France, Canada, the Netherlands, to Indonesia. World Health Organization (WHO) has determined a case of viral infection Monkeypox as a global health emergency. Virus Monkeypox infects humans in the form of structures Intracellular Mature Virion (IMV) via a membrane protein, the E8L protein. The IMV E8L protein has a region that can be recognized and bound to antibodies, called a B cell epitope. B cell epitopes have been extensively studied to find peptide sequences as peptide-based vaccine ingredients. The aim of this study was to find potential B cell epitope sequences for peptide-based vaccine candidates. This research was conducted by in silico through B cell epitope mapping, then several epitope candidates were tested based on several parameters such as antigenicity test, allergenicity test, hydrophobicity test, docking simulation, and analysis of molecular interactions. The result of this research is that epitope potential B cells for peptide vaccine candidates based on antigenicity (0.4274), allergenicity, hydrophobicity (-2.418) tests, and bond energy interaction results docking (-274.0) is epitope with the sequence KKKYSSYYEEAKK. The molecular interaction between the potential epitope and the B cell receptor (ID. 5IFH) is via the amino acids lysinee 1, lysinee 2, and tyrosine 4.
