Dans le but d'assurer un environnement sain pour la faune du mésocosme du Saint-Laurent marin (3 000 m³ d'eau de mer à 10°C), le Biodôme de Montréal utilise un système de dénitrification alimenté au méthanol. Ce système n'offre pas le rendement souhaité, c'est-à-dire une réduction des nitrates à moins de 20mg N/mL. Quatre unités pilotes de 100L, alimentés à même le mésocosme ont été construits afin d'optimiser les paramètres d'opération du système de dénitrification. Ils permettront, entre autres, d'étudier de nouveaux supports à biofilm, de comparer l'opération du système en mode cuvée versus en mode continu, et de calibrer le mode de fluidisation. Des essais préliminaires ont été réalisés en laboratoire pour déterminer la source de carbone et le rapport C/N les plus efficaces. Un milieu synthétique reproduisant l'eau du mésocosme et contenant 300 mg N-NaNO3/L a été inoculé avec la flore microbienne du système de dénitrification. Quatre sources de carbone ont été testées soit le méthanol – le carbone de référence -, l'éthanol, l'acétate de sodium et l'isopropanol. Un mélange méthanol-acétate fut aussi testé. Les essais se sont étalés sur une période d'environ 5 jours. Les paramètres suivants ont été mesurés deux fois par jour : les nitrates, les nitrites, la biomasse bactérienne, le pH et la composition de la flore microbienne. L'activité dénitrifiante a été maximale dans les milieux supplémentés au méthanol dans un rapport C/N égal à 1.5. La dénitrification a été complète (>95%) avec l'acétate et l'éthanol avec des rapports C/N de 2.0 et 1.5 respectivement, mais une accumulation des nitrites fut observée avec l'éthanol. Enfin, le profil de diversité microbienne observé par PCR- DGGE a varié avec la source de carbone. Les résultats obtenus suggèrent d'utiliser le méthanol dans un rapport C/N de 1.5 pour les expérimentations sur les unités pilotes puisque les taux de dénitrification sont les plus élevés et aucune hausse des nitrites n'est observée dans ces conditions. Par contre, il serait intéressant d'étudier le fait que le dénitrificateur du Biodôme doit être alimenté avec un rapport C/N très supérieur au rapport optimal pour être fonctionnel.
