Cytokine responses of chickens to a DNA plasmid encoding a refractile body antigen of Eimeria tenella

Abstract
Effects of doses and routes of delivery of refractile body (SO7) antigen on weights, lesions and cecal interferon gamma (IFN-[gamma]) and interleukin 8 (IL-8) mRNA production in chickens were evaluated. Antigen was ligated into pRc[alpha] plasmid vector; 12.5[mu]g, 25[mu]g, 50[mu]g, or 100[mu]g of plasmid DNA were injected intramuscularly (IM) or 100[mu]g were administered intranasally (IN). Two weeks later half the chickens from each treatment were challenged with 2.5 * 103 oocysts of 'E. tenella', Guelph strain. Weight gains and lesion scores did not differ significantly among challenged and non-challenged groups. Interferon-[gamma] production increased significantly in chickens on day 6 that received 12.5[mu]g, 25[mu]g or 100[mu]g IM, and on day 8 for 25[mu]g, 100[mu]g vector or 100[mu] IN treatments. Interleukin-8 mRNA was elevated on day 2 and 6 in chickens that received 100[mu]g IN and 100[mu]g IM of plasmid DNA, respectively. The pRc[alpha]-SO7 plasmid conferred limited protection against infection with 'E. tenella '.
