Insect resistance associated with different Medicago truncatula ecotypes

Abstract
Insect herbivory may be devastating for a crop. One important pest insect is the beet armyworm, Spodoptera exigua Hübner. This caterpillar causes damage to many different economically important horticultural and agricultural crops. However, in response to herbivore pressure, plants have evolved defenses that can act on the insect by affecting their behaviour, growth and/or mortality; all of which substantially decrease insect pest populations (Karban and Baldwin, 1997). Therefore, to study plant resistance, the model legume Medicago truncatula Gaertn was used. Different M. truncatula ecotypes (A17, A20, DZA045.5, DZA315.16, F83005.5, and TN1.11) were screened for resistance against caterpillars of the beet armyworm. F83005.5 was identified as the most resistance line and TN1.11 as the most susceptible line through in vitro and in vivo choice tests. The constitutive and induced defenses of these two lines were compared, focusing on expression of 6 marker genes (MtPP, MtRFP, MtRCA, MtSTR, MtPR1, MtVSP) and defensive proteins (polyphenol oxidase (PPO) and trypsin inhibitor (TI)). Transcript gene expression was measured by quantitative real-time polymerase chain reaction. PPO and TI were measured spectrophotometrically through enzyme assays. MtPR1 was induced in both the resistant line F83005.5 and susceptible line TN1.11, however, expression was significant higher in F83005.5 than TN1.11. MtRFP expression was induced only in the resistance line F83005.5. Expression of MtRCA was constitutively higher in F83005.5 than in TN1.11. Unlike gene expression, there was no difference in constitutive or induced PPO or TI levels. The knowledge acquired from this project will contribute to understanding legume plant resistance mechanisms against caterpillar pests.
