LA RICOMBINAZIONE COME FONTE PRINCIPALE DI DIVERSITA GENETICA E PATOGENETICA NEI CEPPI DI GAS\nCON SIEROTIPO M89.

Abstract
Con più di 150 sierotipi M Streptococcus pyogenes rappresenta uno dei patogeni umani più versatili.\nI ceppi con sierotipo M89 sono una delle cause più frequenti di faringiti ed infezioni invasive.\nStudi recenti indicano che la prevalenza di ceppi M89 sta aumentando in tutto il mondo.\nIn Italia, il 25% delle infezioni invasive da Streptococcus pyogenes tra il 1994 e il 1996 è stato causato dal sierotipo M89. Successivamente,\nla sua incidenza si è ridotta nel tempo fino ad essere responsabile solo di pochi casi tra il 2003 e il 2005.\nAttraverso uno studio di genomica comparativa abbiamo valutato la diversità genetica del sierotipo M89 e il suo contributo alla patogenesi\ndi Streptococcus pyogenes.\n27 ceppi M89 (Italia = 10, Canada = 1, New Zealand = 1, U.S.A. = 15) sono stati sequenziati mediante tecnologia Illumina. Sono stati\ndeterminati i genomi completi di un ceppo italiano e di un ceppo statunitense. L’identificazione di polimorfismi (SNPs) “genome-wide” ha\npoi permesso di analizzare la variabilità inter- e intra-specie.\nIl genoma del ceppo italiano 11610 (~ 1,70 Mb) è privo di profagi. I ceppi italiani differiscono tra di loro, in media, per circa 30 polimorfismi.\nIl genoma del ceppo statunitense MGAS11027 (~ 1,79 Mb) presenta due profagi codificanti fattori di virulenza. Gli isolati americani\ndifferiscono tra di loro, in media, per circa 500 SNPs. I ceppi italiani si differenziano da quelli statunitensi per circa 2000 polimorfismi. I\npolimorfismi a singolo nucleotide tra i ceppi M89 americani e gli SNPs che differenziano i ceppi statunitensi da quelli italiani non sono\ndistribuiti in modo casuale, ma sono raggruppati in clusters.\nI ceppi M89 presentano un numero di polimorfismi superiore a quello che caratterizza solitamente gli altri sierotipi di Streptococcus\npyogenes. La maggior parte degli SNPs è da attribuire ad eventi di ricombinazione che coinvolgono geni codificanti fattori di virulenza\ncome adesine di superficie, peptidi antimicrobici e proteine ad attività antifagocitaria. Questi risultati dimostrano che la ricombinazione\ngenetica contribuisce significativamente alla diversificazione della capacità patogenetica dei ceppi GAS con sierotipo M89.
