Detection and identification of a novel saxitoxin analogue in scallops (Zygochlamys patagonica)

Abstract
Paralytic shellfish toxins are a group of potent neurotoxins that include saxitoxin and a number of structural analogues that are responsible for numerous poisoning incidents worldwide. The Canadian Food Inspection Agency routinely monitors both domestic and imported shellfish products for paralytic shellfish toxins to ensure the health of consumers in Canada. In 2006, the agency began a real-time comparison study between the official mouse bioassay method for paralytic shellfish toxins and a new, rapid method based on liquid chromatography with post-column oxidation and fluorescence detection. Excellent correlations have been found with thousands of shellfish samples. However, testing of some offshore harvests of scallops (Zygochlamys patagonica) from Argentina showed an apparent discrepancy between method results. High toxicity was observed with the mouse bioassay method but only trace levels of the known paralytic shellfish toxins were detected by liquid chromatography. Upon examination of the chromatograms, an unidentified peak was observed and found to correlate well with mouse bioassay toxicity values. The techniques of fluorescence spectroscopy and liquid chromatography-tandem mass spectrometry were utilized to provide initial structure information on the toxin. Its molecular weight and elemental composition have been measured, and it was observed that the product ion and fluorescence spectra have features similar to those of other saxitoxins. Through a combination of size exclusion and preparative liquid chromatography, the compound was isolated for structure determination. Methodologies used, and the proposed structure of the new toxin will be discussed.
