Latar Belakang: Peningkatan usia harapan hidup terjadi karena peningkatan jumlah lansia, dimana pada lansia telah mengalami penurunan fungsi struktur tubuh. Sehingga lansia mengalami gangguan penyakit degeneratif banyak terjadi yaitu osteoarthritis knee menyebabkan lansia mengalami penurunan dalam aktivitas fungsionalnya sehingga menghambat dalam aktivitas lansia sehari-hari.Tujuan: untuk mengetahui pengaruh latihan isometric otot quadriceps terhadap peningkatan aktivitas fungsional pada penderita osteoarthritis knee, untuk mengetahui pengaruh latihan closed kinetic chain terhadap peningkatan aktivitas fungsional pada penderita osteoarthritis knee, untuk mengetahui perbedaan pengaruh latihan isometric otot quadriceps dan latihan closed kinetic chain terhadap peningkatan aktivitas fungsional pada penderita osteoarthritis knee . Metode: Penelitian ini merupakan experimental dengan pre test and post test two group design sebanyak 14 orang penderita osteoarthritis knee. Sampel dibagi menjadi 2 kelompok yaitu kelompok I mendapat perlakuan latihan isometric otot quadriceps, kelompok II mendapatkan perlakuan latihan closed kinetic chain , keduanya dilakukan 3 kali seminggu selama 4 minggu. Alat ukur dalam penelitian kemampuan fungsional Western Ontario and Mcmaster Universities Osteoarthritis Index (WOMAC) . Uji normalitas data menggunakan uji shapiro wilk-test dan uji homogenitas data menggunakan lavene’s test. Penggunaan Paired samples t-test untuk mengetahui peningkatan aktivitas fungsional penderita osteoarthritis knee pada kelompok I dan II serta menggunakan Independent samples t-test untuk komparabilitas hasil intervensi kelompok I dan II. Hasil: Hasil uji menggunakan Paired samples t-test pada kelompok I p=0,000 (p<0,05) dan pada kelompok II p=0,001 (p<0,05) menunjukkan bahwa kedua perlakuan berpengaruh terhadap peningkatan aktivitas fungsional pada penderita osteoarthritis knee, Sedangkan hasil komparabilitas menggunakan Indepndent samples t-test p=0,046 (p<0,05) menunjukkan bahwa kedua perlakuan memiliki perbedaan pengaruh yang signifikan terhadap peningkatan aktivitas fungsional pada penderita osteoarhritis knee. Kesimpulan: Ada perbedaan pengaruh latihan isometric otot quadriceps dan latihan closed kinetic chain terhadap peningkatan aktivitas fungsional pada penderita osteoarthritis knee. Saran: Peneliti selanjutnya untuk melakukan penelitian lebih spesifik dan dapat mengontrol aktivitas kegiatan dilakukan sampel penelitian sesuai dengan keadaan dapat mempengaruhi osteoarthritis knee.
