Preterm infants are born with underdeveloped lungs and often require assistance from mechanical ventilators to inspire oxygen.Bronchopulmonary Dysplasia (BPD) is a common lung injury in infants exposed to such high oxygen conditions.Encephalopathy of prematurity (EOP) and retinopathy of prematurity (ROP) are cerebral and retinal injuries, respectively, that commonly present alongside BPD.The postnatal exposure to high oxygen concentrations has been mimicked in animals in order to study the resulting injuries.There is currently a lack of animal models investigating the simultaneous effect of mechanical ventilation on the lungs, brain, and retina.The first objective of this thesis was to evaluate the validity of a BPD rat model to reproduce the simultaneous brain and retinal injuries seen in preterm newborns.The second objective of this study was to examine the possible therapeutic effects of stem cell therapy on these injuries.Endothelial colony forming cells (ECFCs) are highly proliferative endothelial progenitors that have been shown to successfully repair the lung structure and microvasculature injuries in the same BPD model used here.This study will evaluate the potential therapeutic effects of ECFCs on brain and retinal injuries.Results show that this BPD rat model reproduces representative brain and retinal injuries of mechanically ventilated preterm newborns with BPD, and treatment with ECFCs seems to show promising antiinflammatory effects in the brain and retina.
