Pyruvate kinase is the enzyme that performs the last, irreversible, rate-limiting reaction to the glycolytic metabolic pathway.Its regulation is critical for the metabolism of the cell.Erythrocytes lack mitochondria for oxidative respiration and thus their sole means of obtaining energy is through glycolysis.The pyruvate kinase isoform found in erythrocytes, designated PK-R and coded by the PKLR gene is responsible for 50% of the ATP production in this cell type.Red cell pyruvate kinase deficiency (PKD) is the most common glycolytic defect causing congenital non-spherocytic hemolytic anemia.The deficiency is caused by mutant variants of the PKLR gene that generate mutant proteins that affect the characteristics of the enzyme.To this date over 200 mutations have been implicated in the disease that cause a range of symptoms from mild anemia to severe transfusion dependent anemia and neonatal death.One such variant of the enzyme is the G1168A point mutation that causes the amino acid D390N substitution.This mutant has already been established to inactivate the enzyme activity.We hypothesized that observing the aspartate residue substituted for an asparagine in a solvent inaccessible location at the A/A' interdomain interface of the protein would disrupt a salt bridge formation and lead to steric clashes that would ultimately lead to a weaker intersubunit interaction and thus render a less stable protein.We completed two different protein stability assays: a cycloheximide assay and S-35 labelling pulse-chase to assess the stability of the mutant relative to the wild-type.We conclude that this mutation has no effect on the stability of the enzyme. RsumLa pyruvate kinase est l'enzyme catalysant l'irrversible et dernire raction limitante du mtabolisme du glucose.Sa rgulation est d'une importance cruciale pour le mtabolisme cellulaire.Les rythrocytes ne possdent pas de mitochondries leur permettant de raliser la respiration oxydative et sont donc limit l'usage de la glycolyse pour gnrer leur nergie.L'isoforme de pyruvate kinase trouve dans les rythrocytes, nomm PK-R et encode par le gne PKLR, est ainsi responsable de la production de 50% de l'ATP gnr par ces cellules.La dficience d'expression de la pyruvate kinase dans les globules rouge est le dfaut glycolytique le plus souvent responsable de l'anmie hmolytique non-sphroide congnitale.Cette dficience est due une mutation dans le gne PKLR qui aboutit la production d'une enzyme non fonctionnelle.A ce jour plus de 200 mutations ont t impliques comme source de cette pathologie, avec des consquences allant d'une anmie lgre une anmie ncessitant des transfusions et pouvant aller jusqu' la mort nonatale.Un de ces variants est due la mutation ponctuelle G1168A et rsulte en la substitution d'acide amin D390N.Il a t tabli que cette mutation inactivait l'enzyme.Nous avons mis l'hypothse que la substitution d'un aspartate par une asparagine l'interface des interdomaines A/A', dans une poche inaccessible aux solvants, prviendrait la formation d'un pont salin, dstabilisant la structure strique de l'enzyme et rendant ainsi la protine moins stable.Nous avons raliser deux essais diffrents, l'un en utilisant du cycloheximide et l'autre en ralisant un pulse-chase marqu au S-35 afin de dterminer la stabilit de ce mutant comparativement la proteine sauvage.Nous concluons que cette mutation n'affecte nullement la stabilit de cette enzyme.
