L'asthme est reconnu comme étant l'une des maladies chroniques les plus répandues dans l'enfance et sa prévalence dans la population générale est de 5 à 10%. On le définit comme étant une maladie complexe, c'est-à-dire sous l'influence de facteurs environnementaux, ainsi que de nombreux gènes, chacun ayant une contribution variable pour chaque individu. Des groupes de chercheurs ont amorcé des études génétiques au sein de plusieurs groupes ethniques afin de documenter les bases moléculaires des différentes formes d'asthme. Ce mémoire s'inscrit dans une vaste étude qui tente d'identifier les gènes de prédisposition à l'asthme au sein d'une population à effet fondateur. Quatre polymorphismes des gènes de FceRI-B, PPAR-a et PPAR-y, identifiés lors d'études antérieures, ont été évalués dans la population du Saguenay-Lac-Saint-Jean ainsi que pour une cohorte familiale de la région métropolitaine de Québec, en fonction du phénotype asthme. Les résultats obtenus à l'aide du test de déséquilibre de liaison (TDT test) ont montré qu'il n'y a aucune association entre le variant E237G-FceRI-P et l'asthme et les conditions reliées à l'asthme. Aucun participant à l'étude ne possédait le variant I181L-FceRI-p. Cette étude a permis de démontrer que la transmission combinée des variants L162V-PPAR-a et P12A-PPAR-y semble présenter un effet protecteur en ce qui concerne l'atopie. Cette observation n'a toutefois pas été confirmée au sein de la cohorte familiale originaire du Québec métropolitain. \n
