The rising global population and increasing demand for plant protein foods warrant the utilization of existing protein resources. The value-addition to protein-rich by-products becomes necessary to meet the burgeoning demand. Corn distillers solubles (CDS) is an underutilized by-product from bioethanol industries in Canada along with 16000 MT annual production from Integrated Grain Processors Co-operative Inc., Ontario. Protein fraction (PF) extracted through two-step sedimentation from CDS containing corn and yeast proteins on trypsin hydrolysis demonstrated potential for protein hydrolysates. Analytical pyrolysis revealed aggregation, cross-linking, and glycation characteristics of PF proteins. The total nitrogen (protein) compounds were 12, 21, 16, and 23% through fast pyrolysis of PF, hydrolysate created via alcalase (PH-A), papain (PH-P) and trypsin (PH-T), respectively. Amines & amides from PF, N-heterocyclics from PH-A, nitriles from PH-P, and amines & amides from PH-T were perceived as the greatest categories. Optimization of alcalase hydrolysis of protein concentrate (PC) from PF provided optimal E:S ratio and incubation time to create antioxidant PHs. Kinetic model for PC using alcalase was developed to find critical concentrations of enzyme and substrate along with degree of hydrolysis. Furthermore, PH-A among one-step [A, T, P, and flavourzyme (F)] and PH-AF among two-step enzymolysis of PC [AF, FA, AT, TA, FT, TF] were optimal PHs with angiotensin-converting enzyme (ACE) inhibition and dipeptidyl peptidase-4 (DPP4) inhibitions. The key antioxidant peptides, FLPF, VALPL, LLPF, QQPIIGGA, LPPYLSPA, SNIPLSPL, NPILQPY, NIIPSPI and anti-ACE peptides, GALLLPF (corn), RLLPF (yeast), and HGDWPLPT (corn/yeast) were detected in PH-A. Besides, peptides like FDFFDNIN and WNGPPGVF from the PH-A; DPANLPW and TPPFHLPPP from PH-F inhibited ACE more effectively. Similarly, WNGPPGVF from PH-A; WPLPPFG and LPPYLPS from PH-P; TPPFHLPPP and DPANLPWG from PH-F had greater inhibition against DPP4. The PH-AF comprised APLA, PLFP, LFLP, LPPYL, PLYPLP, NDWHTGPL, LPPYLPS, GSPFLGQ, SWQQPIVGG capable of inhibiting ACE and DPP4. Investigation using bioinformatic tools demonstrated the potential of the peptides from CDS as non-toxic, non-allergenic and highly bioactive resources. Furthermore, studies using mass balance analysis for PH-A and PH-AF has revealed the opportunities for scale-up of the developed processes. The outcome of this study demonstrates the opportunities for developing valorized products from CDS for feed, food, nutraceutical, and pharmaceutical industries.
