A Brucella canis, causadora da brucelose, é um patógeno listado pelo Organização Mundial da Saúde Animal (WOAH) como uma das zoonoses emergentes negligenciadas na atualidade. É responsável por perdas econômicas, assim como problemas de saúde pública. Estima-se que haja meio milhão de casos de brucelose ao ano no mundo, sendo destes, 1% causados pela B. canis. Apesar disso, ainda há poucos estudos acerca do perfil genômico e quanto ao perfil de susceptibilidade a antimicrobianos para esta bactéria. Por isso, esse trabalho tem como objetivo realizar a caracterização genômica e fenotípica de 19 isolados de B. canis isolados de caninos do sul do Brasil entre os anos de 1995 a 2015. Para isso, realizou-se análises in sílico e in vitro. Para a caracterização genômica e filogenética, realizou-se a montagem híbrida das reads brutas geradas através de dois sequenciadores de última geração distintos. Para os testes de susceptibilidade in vitro, determinou-se a concentração inibitória mínima (CIM) e concentração bactericida mínima (CBM) dos isolados frente a quatro antimicrobianos. Através das análises genômicas, detectou-se a presença de dois genes de resistência a antimicrobianos em todos os isolados, denominados DNA gyrase subunit A (gyrA) e Alanyl phosphatidylglycerol synthase (A-PGS). Além disso, as análises filogenéticas demonstraram o predomínio do sequence type 20 (ST20) entre as cepas desse estudo, assim como a delimitação de duas linhagens de B. canis distintas baseada em SNPs. Quanto ao perfil de susceptibilidade a antimicrobianos, os isolados do presente estudo demonstraram um perfil semelhante entre eles, onde a gentamicina foi a droga mais eficaz nos ensaios in vitro. Por fim, os resultados obtidos através desse trabalho contribuem com informações acerca do perfil genômico e filogenético de isolados de B. canis do sul do Brasil, possibilitando novas perspectivas de tratamento e prevenção para a brucelose canina.
