Investigating the lipidome of the lateral ventricle choroid plexus during ageing and across sex

Abstract
During ageing, the brain's lipid composition and the cerebrospinal fluid (CSF) secreted by the choroid plexus (ChP) undergo significant modifications. The ChP, an epithelial tissue in each brain ventricle, experiences a decline in its functions, including protein secretion and CSF production. A crucial gap in understanding the ChP lies in its lipid composition and the potential repercussions of these changes on ChP physiology during ageing. This study hypothesised that the lipidome of the lateral ventricle choroid plexus (LVCP) is modulated during ageing, potentially generating pro-inflammatory mediators. The objective was to perform quantitative lipidomics on mouse LVCP during ageing and investigate whether pro-inflammatory lipid mediators increase as the mice age. LVCP and cortex tissues from mice aged 6, 12, 18, and 24 months (n = 5/age/sex) were collected from C57BL/6 mice of the Quebec Network for Research on Ageing. These tissues were analysed using liquid chromatography coupled with tandem mass spectrometry, and statistical analyses were conducted using the Lipidr software package and univariate statistical methods. During ageing, fifteen lipids in the LVCP showed significant changes: eight on a phosphatidylethanolamine (PE) backbone and six on a phosphatidylcholine (PC) backbone. Four lipids containing eicosapentaenoic acid were more abundant in 6-month-old mice than in 18-month-old mice, indicating a decrease in precursors of anti-inflammatory lipid mediators during ageing. No significant changes were observed in the lipids of 12-month-old mice compared to other ages. Phospholipids containing eicosapentaenoic acid were the most modified during ageing, with all significantly changed phospholipid species having lower concentrations in 18-month-old mice. The LVCP of males and females exhibited thirty-three lipids with distinctive profiles between sexes, notably in sphingomyelin, which were less concentrated in males. Additionally, there was a sex-by-age interaction for seven lipid mediators, six derived from linoleic acid. Contrary to the hypothesis, the lipidome of the LVCP did not favour a pro-inflammatory lipid profile or mediator profile during ageing, suggesting that the LVCP does not contribute to the synthesis of pro-inflammatory lipid mediators released in the brain during ageing.
