Abstract
Crocanthemum canadense(L.) Michx., commonly known as Rockrose, is an endangered plant native to northeastern North America. Rockrose has a dual breeding system producing two types of flowers in different locations on the same plant in the same growing season: chasmogamous (cross-fertilizing) flowers and cleistogamous (self-fertilizing) flowers. Yorke et al. (2011) showed Nova Scotia populations of Rockrose had the lowest genetic variation of those examined in its northeastern limit. These results left the question of whether an underlying reason exists beyond disjunction, such as reproductive influences, for the reduced genetic variation in Nova Scotia. Additionally, several new populations were discovered in Annapolis Valley along Highway 201 and on the Greenwood Air Force Base. Amplified fragment length polymorphisms (AFLPs) were used to measure and compare genetic diversity of plants of known pedigree and individuals belonging to newly discovered populations. Data were examined with analysis of molecular variance (AMOVA), principal coordinate analysis (PCoA), and a neighbour-joining tree to illustrate relationships among individuals and populations. It was determined no significant genetic variance existed between plants raised from cleistogamous or chasmogamous seeds. Very little genetic diversity was found between new populations examined, and a comparison against old populations also revealed very little genetic variation. Low population variance is likely due to continual inbreeding and low rates of cross-fertilization due to isolated, small populationspresent in Nova Scotia. Implications from this research may inform future conservation decisions. Potential breeding projects can use seeds produced by either cleistogamous or chasmogamous flowers without compromising the genetic integrity of Nova Scotia populations.
