Abstract
Genomic DNA clones used in the construction of all 3 mouse lines were isolated from the 129/Svj mouse genomic DNA BAC library (Invitrogen). Genomic DNA fragments of interest were subcloned in pBluescript and targeting vectors were constructed based on information from mouse genomic DNA databases. PICK1 KO and GluR2∆7 KI mice were generated as previously detailed (Gardner et al., 2005). GluR2 K882A KI mice were generated using a strategy very similar to that for GluR2∆7 KI mice. Targeting vectors were constructed with thymidine kinase genes for negative selection and neomycin resistance cassettes for positive selection. Mutagenesis was performed in pBluescript using the QuikChange method (Stratagene) and verified by sequence analysis. Linearized targeting vectors were electroporated into R1 embryonic stem (ES) cells (Dr. A. Nagy, Mount Sinai Hospital, Toronto, Canada) and homologous recombinants were isolated by positive and negative selection with G418 and gancyclovir, respectively.
