Abstract

Based on H3 numbering (S1), cDNA corresponding to residues 11–329 (HA1) and 1–176 (HA2) of the ectodomain of the hemagglutinin (HA) from A/Vietnam/1203/2004 (Viet04; Accession No. AY651334 & AY818135) (S2), A/Duck Alberta/35/1976 (H1_Duck; Accession No. AF091309 & D10477) and A/Duck/Singapore/Q-F119-3/1997 (H5-Duck; Accession No. AF303057) was cloned into the baculovirus transfer vector, pAcGP67-A (BD Biosciences Pharmingen, San Jose, CA). In a similar fashion to the construct used for expression of the 1918 influenza virus HA for structural studies (S3), the construct also incorporated a C-terminal thrombin cleavage site, a "foldon " sequence (S4) and a hexa His-tag at the extreme C terminus of the construct to enable protein purification. Once sequence verified, the plasmid was used for mutation studies.
