L’infertilité affecte un couple sur six dans le monde, avec des facteurs masculins contribuant à plus de 50% de cas, dont 34% restent idiopathiques. Malgré cela, les outils de diagnostic dans les cliniques de fertilité sont restés stagnants pendant des décennies, ne parvenant pas à évaluer les fonctions cruciales du sperme telles que la capacitation. La capacitation est cruciale pour la fertilisation, impliquant des changements biochimiques tels que la production réactive des espèces de l’oxygène (ROS), la phosphorylation, les altérations de la composition lipidique et le flux de cholestérol. On sait déjà que les sphingolipides bioactifs, tels que S1P, sont fondamentaux pour réguler de nombreux domaines et processus physiologiques. La recherche détaillée dans la première partie de cette thèse explore le rôle des sphingolipides dans la capacitation du sperme humain. Nous avons constaté que S1P engage son récepteur S1PR1 couplé à Gi pour arbitrer la compétence de fertilisation, favorisant ainsi l’activation de la voie PI3K-AKT et la production d’oxyde nitrique (NO•). L’inhibition de la synthèse d’oxyde nitrique a altéré la capacité, alors que la sphingosine (Sph) et le céramide (Cer) induisait la production d’anion de superoxyde (O2•−), qui a été bloquée par la dismutase de superoxyde (GAZON). PKR, CERK, et PKC transloquaient et activent la kinase de sphingosine 1 (SphK1), nécessaire pour la production de S1P pendant la capacitation. Nos études jettent les bases de l’examen du profil lipidique des mâles infertiles, car les écarts potentiels peuvent affecter la capacité fonctionnelle des spermatozoïdes à atteindre le potentiel de fécondation. La deuxième étude étudie comment la dyslipidémie causée par l’obésité altère la fertilité masculine. Dyslipidémie, caractérisé par des profils anormaux de lipide (par exemple, triglycérides élevés et bas HDL), affecte la qualité de sperme en perturbant l’homéostasie de lipide et en déclenchant l’effort oxydant. Le flux de cholestérol, un événement clé dans la capacitation, modifie la composition de lipide et la fluidité de membrane. Cependant, l’interaction complexe entre le flux de cholestérol et les modifications capacitation-associées dans les spermatozoïdes humains demeure mal comprise. Utilisation de méthyl-β-cyclodextrine (MβCD) pour induire le flux de cholestérol, par la signalisation redox, la capacité améliorée, la motilité hyperactive et la réaction acrosome. Cependant, métabolisme des lipides dérégulé, avec le cholestérol-sulfate élevé, la Cer, et la Sph, a altéré la capacité et a induit l’effort et les dommages oxydants. Ces résultats suggèrent que les perturbations du métabolisme des lipides, en particulier dans l’obésité, altèrent la capacité de sperme et le potentiel de fertilisation, fournissant de nouvelles perspicacités dans les mécanismes masculins d’infertilité
