Le carcinome basocellulaire (CBC) est la forme de cancer de la peau la plus fréquente, mais ses caractéristiques moléculaires demeurent encore partiellement comprises, en particulier en ce qui concerne les phénotypes précoces à haut risque. Ce mémoire de maîtrise présente une étude génomique et transcriptomique intégrée du CBC, s’appuyant sur les plateformes de séquençage à courtes lectures (Illumina) et à longues lectures (Oxford Nanopore) pour l’ARN et l’ADN. Nous avons profilé les mutations somatiques, les gènes différentiellement exprimés (DEGs) et les fusions géniques dans des échantillons de CBC et de tissus normaux appariés, identifiant des signatures moléculaires distinctes associées aux sous-types tumoraux. L’analyse génomique a révélé des mutations dans des gènes conducteurs classiques du cancer, tandis que l’analyse transcriptomique a mis en évidence une dérégulation des voies liées à la matrice extracellulaire et au métabolisme lipidique. L’analyse spécifique aux sous-types a également révélé un enrichissement des voies de migration immunitaire dans les sous-types agressifs. Des fusions géniques nouvelles et complexes ont été identifiées, y compris celles impliquant des ARN non codants et des gènes associés au cancer. L’intégration des données issues des séquençages Illumina (courtes lectures) et Nanopore (longues lectures) a permis d’obtenir des perspectives complémentaires et convergentes, soulignant l’intérêt d’utiliser ces deux technologies pour mieux caractériser le paysage moléculaire complet du CBC. Ces résultats enrichissent notre compréhension des altérations génétiques et transcriptionnelles du CBC et mettent en lumière des biomarqueurs candidats présentant un potentiel pour l’oncologie de précision dans le cadre des cancers cutanés non mélanocytaires
