Abstract
In order to consider an assay “validated” for a gene it must be able to distinguish between WT-like variants and NULL-like variants consistently and reliably, and preferably in a pooled format. This protocol outlines how to generate variants selected that are known to cause disease (clinically annotated pathogenic or dysfunctional), or to otherwise have a disruptive effect on protein structure or gene function, and a few variants that are known to not have an effect (i.e. clinically annotated benign or behave like the WT gene).
