Investigating the Physiologic Role of the Stromal Factor CXCL12 in Murine Kidney Development

Abstract
In the mammalian kidney, Forkhead box D1+ (FOXD1+) stromal cells engage inreciprocal signaling with adjacent nephrogenic and ureteric tissues, thus controlling their embryonic development. Published work from our lab demonstrated that constitutive activation of Hedgehog signaling within the Foxd1+ embryonic stroma led to renal hypodysplasia characterized by reduced kidney size, fewer nephrons and ureteric branches, and expanded medullary cellularity. Single cell RNA sequencing of mutant kidneys revealed an upregulation of C-X-C motif chemokine ligand 12 (CXCL12) in the stromal lineage. To understand the role stromal CXCL12 signaling plays in kidney development, I generated and characterized stromal Cxcl12-deficient kidneys using a Foxd1Cre; Cxcl12loxP/loxP mouse strain. Mutant kidneys exhibited renal hypoplasia characterized by decreased kidney size, and fewer nephrons and ureteric branches. Bulk RNA sequencing of mutant kidneys identified the Wingless-type MMTV integration site family member (WNT) signaling inhibitor DACT1 to be a possible downstream effector of stromal CXCL12 signaling. Taken together, these data suggest that stromal CXCL12 acts downstream of Hedgehog (HH) signaling and acts in a cell non-autonomous fashion to regulate nephrogenic and ureteric lineage development.
