Abstract
Previous research has established that HIV-1 gene expression is sensitive to modulations made to trans-acting host factors Serine-Arginine (SR) protein. Cdc2-like kinases (CLK1 - 4) that phosphorylate SR proteins, exhibit >89% sequence similarity within their kinase domain. However, when overexpressed, HIV-1 Gag expression is enhanced by CLK1, reduced by CLK2, while CLK3 and -4 had no significant effect. This thesis explores the roles of the CLK domains in regulating HIV-1 gene expression. Analysis of CLK chimeras revealed that the RS domain of CLK1 and 2 influenced activity, dependent on the kinase domain. Furthermore, deletion analysis of the CLK RS domain revealed functional differences; mutants CLK1-∆2 and CLK1-∆3 had opposing effects on HIV-1 gene expression, in contrast, the entire RS domain was required for CLK2 function. Utilizing miniTurbo-CLK fusions proteins and TMT-MS, quantitative comparisons of CLK1-WT, CLK1-∆2, CLK1-∆3 and CLK2-WT interactomes identified a small subset of interactions that preferentially associated with the CLKs, providing insight into the observed functional differences.
