Abstract
Screening approaches have identified many peptide-based biologics with demonstrable in vivo efficacy. However, clinical translatability is impeded by outstanding challenges in target delivery. Such macromolecules are often large and complex and do not easily diffuse across membranes, and thus require co-delivery with carrier machinery capable of selective transport to the desired tissues. Some of the most attractive avenues are bacterial exotoxin-based platforms which appropriate specific proteinaceous domains for bespoke surface receptor recognition and cytosolic translocation. A complete profile of receptors involved during uptake of these effectors could thus expand the available tissue repertoire of important therapeutic molecules. To this end, this thesis leverages genome-wide loss of function screens to discover surface factors responsible for the cellular invasion of two modified exotoxin constructs.
