Abstract
G protein-coupled receptors (GPCRs) are the largest family of membrane proteins and mediate signaling for a host of neurotransmitters, hormones and other stimuli from light to odorants. Despite their ubiquity and importance, their mechanism of action remains unknown. That uncertainty derives in part from uncertainty over their oligomeric status and the origin of a characteristic, efficacy-correlated heterogeneity revealed in the binding of agonists. If GPCRs signal as monomers, the heterogeneity could be induced by the G protein; if they signal as oligomers, it can be attributed to a mix of cooperativity and asymmetry among orthosteric sites within the oligomeric array.
