Investigating the Roles of Truncated Kinases in Oncogenic Signal Rewiring

Abstract
Cellular responses to extracellular signals are an integral aspect of homeostasis and are frequently controlled by kinases that regulate the functions of their substrate proteins. Thus, kinase activity is tightly regulated, allowing signal transmission to occur at the appropriate time. The dysregulation of kinases, particularly, deletions of regulatory regions, is frequently linked with cancers. I hypothesize that kinase truncations rely on specific sets of protein associations to mediate oncogenesis and that these are potential therapeutic targets. Using BioID and live imaging, I systematically characterized the protein associations of truncated kinases originating from various cancer types. BioID, a proximity-dependent biotinylation proteomics approach can uncover the protein associations around a protein of interest. Importantly, my BioID results are validated by the live fluorescent microscopy data. Overall, the data indicate that kinase truncations can rewire signalling pathways and may lead to the identification of new potential targets for novel therapies.
