Quantifying Osteosarcopenia Progression in a Rodent Model of Mixed Osteoblastic-Osteolytic Bone Metastases

Abstract
Growing evidence shows a link between sarcopenia and osteopenia in many diseases, including cancer. Systemic chemotherapy treatments with docetaxel (DTX) and bisphosphonates [zoledronic acid (ZA)] may impact muscle and bone. This study aims to quantify early changes to the psoas muscle secondary to prostate cancer and its treatment. Athymic, 6-week-old male rats (n=12) received intra-cardiac ACE-1 prostate cancer cell injections (control n=18). Grip strength, in-vivo µMR, and µCT were performed serially to assess muscle function, volume, and attenuation. Rats were randomized into DTX, ZA, or untreated groups. Tumor-bearing rats treated with ZA showed significantly less psoas volume increase compared to healthy controls. DTX-treated rats had significantly less decline in psoas attenuation and lower intramuscular fat content, compared to tumour-bearing untreated animals. Attenuation changes correlated with functional strength, while muscle volume showed weaker or no correlations. These biomarkers may be used to evaluate disease progression, leading to earlier intervention of osteosarcopenia.
