Abstract

The total synthesis of callystatin A 128 and C 19- epi-callystatin 123 was achieved. The convergent strategy relied on the preparation of three fragments, the C1-6 lactone precursor 90 (chapter 5), the C7-12 fragment 108 (chapter 6) and the C13-22 polypropionate fragments 66 and 79 (chapter 4). The fragments were assembled using E-selective tributylphosphonium Wittig olefination reactions (chapter 7). The stereochemistry of the polypropionate fragments 66 (callystatin A) and 79 (C19- epi-callystatin A) was secured using an enantiomerically pure [3.2.1]oxabicycle which had been previously described in the literature. The further development and application of this [3.2.1]oxabicycle methodology comprises most of work described in this thesis and is covered in chapters 3 and 4.
