Abstract
Post-translational modifications with ubiquitin chains represent a dynamic, reversible mechanism for regulating protein fate and signaling pathways. The signaling pathways governed by linear ubiquitin (Met1-Ub) chains and the associated biological roles are still emerging. In vertebrates, Met1-Ub chains are built by the Linear Ubiquitin Assembly Complex (LUBAC), and its catalytic subunit HOIP. OTU deubiquitinase With Linear Linkage Specificity (OTULIN), also called GUMBY or FAM105B, selectively hydrolyzes Met1-Ub chains and directly associates with HOIP. Here, I describe novel roles for OTULIN in PCP signaling and Parkin-mediated mitophagy. I show that in PCP signaling, OTULIN protein interactions can direct LUBAC recruitment, as well as Met1-Ub target and pathway choice. Met1-Ub, which has been previously shown to control protein activity or stability, can also direct the surface presentation of transmembrane proteins. I show that OTULIN is involved in Parkin recruitment to mitochondria. In addition, HOIP recruits Parkin to mitochondria and co-localizes with Parkin at mitochondria. My work contributes to the short list of known LUBAC targets that are modified with Met1-Ub chains. My research also expands the role of OTULIN in Parkin-dependent mitophagy, contributing to unexplored cellular functions of OTULIN and LUBAC.
