[68Ga]-Albumin-PET and [18F]-FDG-PET in the monitoring of left ventricular function in murine models of ischemic and dilated cardiomyopathy: Comparison with cardiac MRI

Abstract
1651 Objectives To evaluate left ventricular functional parameters in healthy mice (HC) as well as in murine models of ischemic (ICM) and dilative cardiomyopathy (DCM) with the novel blood pool tracer (BP) PET tracer [68Ga]-albumin (AlbBP) and with [18F]-FDG-PET. Methods Myocardial infarction (MI) was induced in mice (n=7) by surgical occlusion of the left anterior descending artery. Mice were examined 4 weeks after MI by ECG-gated PET and MRI. To obtain a murine model of DCM mice (n=4) were infected with Coxsackie virus B3. Our DCM mice were examined by ECG-gated PET and cine MRI approximately 16 weeks after infection. Healthy, aged-matched mice (n=4) served as controls. Cine-MRI examination performed on a clinical 1.5T MRI provided the reference standard measurement. The corresponding left ventricular function parameters (EDV, ESV und EF) were calculated and presented as mean ± standard deviation of the mixed group (ICM, DCM, HC). Results Cine MRI results for EDV, ESV and EF were: 125±80µl, 96±77µl, 33±21%. LV volumes and the LVEF as measured by FDG-PET correlated significantly with the MRI results. LV volumes were systematically underestimated by FDG-PET such that EF measurements exceeded results obtained by MRI: EDV 72±37µl (R=0.90), ESV 42±32µl (R=0.88), EF 48±15% (R=0.92). AlbBP significantly overestimated the magnitudes of EDV and ESV whereas the EF matched closely to cine-MRI results with high correlations found between the LV functional parameters: EDV 181±86µl (R=0.98), ESV 136±92µl (R=0.99), EF 31±16% (R=0.99). Conclusions Measuring LV function in mice with AlbBP-PET and FDG-PET is feasible and showed a high correlation compared to cine-MRI. Especially in the ICM-model AlbBP-PET indicates its superiority as compared to FDG-PET.
