Azetukalner is a novel, potent KV7 potassium channel opener with phase 3 trials ongoing for focal onset seizures (FOS), primary generalized tonic-clonic seizures, and planned for major depressive disorder (MDD). In the X-TOLE, phase 2b study in 325 adults with ≥4 focal seizures per month on 1–3 antiseizure medications (ASMs), azetukalner (10, 20, and 25 mg once-daily [QD] with food and no titration period) demonstrated dose-dependent reduction in FOS frequency: 33.2% (10 mg, P=0.04), 46.4% (20 mg, P<0.001), and 52.8% (25 mg, P<0.001) compared to 18.2% for placebo, and early onset of efficacy occurring within 1 week (10 mg P<0.05; 20 mg P<0.05; 25 mg P<0.001 vs placebo).
